Title: WBC diluting fluid destroys RBC but not WBCs !!??

Hello,
While  performing the the total count of WBC , one needs to add a particular volume of WBC diluting fluid to a particular volume of blood . Interestingly, the WBC diluting fluid containing acetic acid (probably 2%) and Gentian violet ( to stain the nucleus of the WBCs)lyses the RBCs only ??!!

Can anyone explain this ? genegenie, there is no genetics involved, even then you should try this !!

Not dull at all.  Medical abbreviations are a foreign abbreviation.  Worse yet are the abbreviations and initials they use in aviation.  They're ridiculous.  Almost nothing has a full name.  I felt pretty dull before I learned all of those.

I was a Navy man....we had acronyms for everything. I almost forgot how to speak english. Now, medical is a whole world of latin as well as acronyms for the latin. I think sometimes it's just made up stuff. I know aviation would be tough as well.
But c'mon....WBC and RBC? I should've known that. I'm just going to beat myself up awhile over that one.

I just want to know how it all got twisted into some sort of perversion. Was it the scrubs comment? I just like to think that I have a strong libido. Yeah, that's it...a strong libido.

As far as the topic, I'll take a stab and say that RBC's have a chemical make up (perhaps protiens) that are affected differently than the WBCs.

Just a thought.

You think with the diluting fluid, perhaps the chemicals react differently with the red cells because of the hemoglobin (iron) content of the cells?

I don't know much biology, but I know that the structure of WBC's is very different from the RBC's.

Lysis - does that mean a loosening of the red blood cells?  If so, Gentian (which is used as a circulatory aid) would theoretically be useful for RBC and cold agglutination.

Do WBCs ever stick together like RBCs sometimes do?

Lysis means gradual dissolution or destruction.  I've never heard of anyone getting Gentian IV for cold agglutination.  That's an interesting thought.  I don't think white cells agglutinate like red cells, at least I don't recall hearing of it at all.  With fibrinogen they will, but I think you're asking like an agglutination syndrome.

Curiosity got the better of me and I looked this one up.[:I]
Apparently the 2% acetic acid is hypotonic to the RBC causing it to swell and lyse.

I'm curious now about whether WBCs can agglutinate. Does this occur in some disease states? Or maybe some infections????

Yes, but that still doesn't explain why the WBC's don't blow up...is 2% acetate not hypotonic to them then ?

Actually both the RBC's and WBC's are lysed.  However, unlike RBC's, WBC's have nuclei and those are what are counted under a microscope or even in a Coulter counter.

the main aim is to concentrate the WBC in a given sampl of blood.. so v ve to lyse the RBCs.. and thats wat dilutin fluid does..

I just fiinished senior level microbiology at UNCW, and we used a premixed solution labeled wright's stain, then DI water.  Does that also contain acetic acid??  None of the cells were lysed for our WBC counts?
