Title: Effect of pH on DNA backbone? (biochemistry)
Post by: Patty on 19/09/2006 02:36:49
Can hydrogen ion concentration (low pH) break the DNA backbone? Can it do it in a sequence-specific manner under any conditions?

I've found a few articles that indicate this might happen, but nothing conclusive. So I'm still wondering if anyone has any knowledge about this.

I believe that the seqience specificty we see may be the resul;t of inverted repeats in the DNA< causing the DNA to assume a cruciform conformation with unpaired bases in the loops. Still, we're at a bit of a loss to explain why the DNA is cleaving in our samples.

Extracting the patented technique is not so easy .
