I think the most likely explanation is that addition of significant quantities of organic solvents (here they add 50 mL urine to 200 mL acetone) will change the solubility of all the originally dissolved species, notably decreasing the solubility of salts, and highly polar molecules (like urea), which could crash out of solution if in high enough concentration initially. (I don't about acetone, but for acetonitrile, the solubility of urea is about 1/20th that of water.) This treatment could also precipitate proteins (not sure if it would denature them or not...), many of which have been studied as diagnostic markers of various diseases.

Human Chorionic Gonadotropin (hCG) is only produced by a small minority of cancer types,
so even assuming the DIY purification process was carried out correctly, in most cases it would not be a useful screening/monitoring test for cancer.
