Adult human males produce sperm on a continuous basis in the testes. There is a temporary store for sperm in the epididymus (http://en.wikipedia.org/wiki/Epididymus#Role_in_storage_of_sperm_and_ejaculant), which also allows the sperm to mature over a period of 2-3 months.
When the temporary store is exhausted, sperm count drops and fertility is reduced. You must wait until more sperm matures.
Semen consists of other fluids produced by a variety of glands (http://en.wikipedia.org/wiki/Semen#Composition_of_human_semen), including the prostate and seminal vesicles.
The rate of semen production is affected by a number of factors including age, hormones, sexual activity and stress. In many species semen production is highly seasonal.
